You should look into the log file.

! pdflatex: major issue: So far, you have not checked for MiKTeX updates.

It seems you may need an update.

tinytex is the easiest way to get started knitting to PDF, unless you have a specific need which necessitates a full LaTeX install, you will generally have far fewer headaches.
