Permission problem

I am trying to use metaDigitize in Rstudio.
After I read the file, it asks for selection 1, 2 or 3. But as soon as I type 1, which I want, it shows following message.

This is what I get after putting these commands.

img1 = metaDigitise(file.choose())

Do you want to...

1: Process new images
2: Import existing data
3: Edit existing data

Selection: 1
Congratulations! Looks like you have finished digitising all figures in this directory.
In addition: Warning message:
In dir.create(cal_dir) :
** cannot create dir 'C:\Users\user\Desktop\IL-6 MA\figure2.png\caldat', reason 'No such file or directory'**
